First report and molecular confirmation of Potyvirus phaseovulgaris associated with yellow mosaic on cowpea Vigna unguiculata in Iraq

Potyvirus phaseovulgaris also known as Bean common mosaic virus (BCMV) was investigated in cowpea samples collected from a field located at Al-Jadriya/Baghdad. Sampling included leaves collected from symptomatic plants (n=16). RNA was extracted from leaf samples and exposed to cDNA synthesis. The presence of BCMV was detected using Nested Revers Transcriptase Polymerase Chain Reaction (N-RT-PCR) and species-specific primer sets. Gel electrophoresis pattern revealed, nested primers designed could targeted BCMV when the resultant expected size DNA fragments of ~500 bp were amplified from 2 out of 16 samples only indicating they could be BCMV infected. Nucleotide sequence comparison confirmed BCMV detection in samples when sequences of 500 bp DNA fragments amplified shared maximum 98% nucleotide identity with the equivalent sequence of a Chinese isolate (MN786956) retrieved from the National Center for Biotechnology Information NCBI data base. Our data suggests the first confirmation of BCMV on cowpea in Iraq based on molecular approaches
